O-21: Differential Expression and Epigenetic Pattern of HOX Family Genes in Cumulus Cells of Mature MII Oocytes from Patients with Polycystic Ovary Syndrome

چکیده مقاله:
Background Ovarian tissue cryopreservation represents a promising strategy to preserve the ovarian function in cancer patients. It is usually performed by slow freezing/rapid thawing (SF/RT). Recent studies emphasize an ultrarapid cryopreservation procedure, vitrification/warming (V/W), since it might prevent damages due to ice crystal formation. Comparative studies between the cryopreservation procedures are primarily based on morphological evaluation of ovarian tissue. This study aims to investigate the bioenergy/oxidative status using Confocal Laser Scanning Microscopy (CLSM) of ovarian tissue cryopreserved by SF/RT and V/W, in association with a morpho-ultrastructural analysis by Light (LM) and Transmission Electron Microscopy (TEM). MaterialsAndMethods Six ovarian biopsies of consent cancer patients were cryopreserved by SF/RT and V/W. Fresh and cryopreserved tissues were processed for CLSM using MitoTracker Orange CMTM Ros (mitochondria activity) and DCHFDA (intracellular reactive oxygen species levels-ROS) fluorescent probes and for ematossilin/eosin staining (LM) and TEM. Mitochondria activity and ROS levels of fresh, SF/ RT and V/W samples were compared by ANOVA. Results Bioenergy/oxidative status resulted significantly different in fresh and cryopreserved ovarian tissues. Fresh samples presented higher mitochondria activity and ROS levels than SF/RT (P=0.002 and P

O-29: Differences in The Transcriptional Profiles of Human Cumulus Cells Isolated From MI and MII Oocytes of Patients with Polycystic Ovary Syndrome
Background: Polycystic ovary syndrome (PCOS) is a common endocrine and metabolic disorder in women. The abnormalities of endocrine and intra-ovarian paracrine interactions may change the microenvironment for oocyte development during the folliculogenesis process and reduce the developmental competence of oocytes in PCOS patients who are suffering from anovulatory infertility and pregnancy loss....
متن کاملEpigenetic modification of long interspersed elements-1 in cumulus cells of mature and immature oocytes from patients with polycystic ovary syndrome
OBJECTIVE The long interspersed elements (LINE-1, L1s) are a group of genetic elements found in large numbers in the human genome that can translate into phenotype by controlling genes. Growing evidence supports the role of epigenetic in polycystic ovary syndrome (PCOS). The purpose of this study is to evaluate the DNA methylation levels in LINE-1 in a tissue-specific manner using cumulus cells...
متن کاملDifferential Expression of Circulating miRNA-103 in Women with and without Polycystic Ovary Syndrome
Background and Aims: Polycystic ovary syndrome (PCOS) has a largely unknown etiology and is a common heterogeneous endocrine disorder in women of reproductive age, characterized by polycystic ovaries, hyperandrogenism, insulin resistance, and chronic anovulation. MicroRNAs (miRNAs) are small, noncoding RNA sequences that negatively regulate gene expression at the post-transcriptional level. miR...
متن کاملMajor Epigenetic Changes in Polycystic Ovary Syndrome
Introduction: Polycystic Ovary Syndrome (PCOS) is one of the most common causes of female infertility. The etiology of this condition is complex. Various genetic and environmental factors have been identified in the pathogenesis of PCOS. The prevalence of this syndrome among Iranian females has been reported between 7.1% to 14.6%. Common symptoms include irregular menstruation, infertility, ova...
متن کاملGene expression microarray profiles of cumulus cells in lean and overweight-obese polycystic ovary syndrome patients.
The aim of this work was to study gene expression patterns of cultured cumulus cells from lean and overweight-obese polycystic ovary syndrome (PCOS) patients using genome-wide oligonucleotide microarray. The study included 25 patients undergoing in vitro fertilization and intra-cytoplasmic sperm injection: 12 diagnosed with PCOS and 13 matching controls. Each of the groups was subdivided into l...
متن کاملO-28: Role of HOX Family Related Genes in Pain Generation of Endometriosis Patients
Background Endometriosis is a common gynecological disease, can cause severe pelvic pain. Studies demonstrated the presence of sensory nerve fibers in endometrium of endometriosis patient. Nevertheless, no information is available on mechanisms of sensory nerve formation in eutopic or ectopic lesions. Since HOX genes have important roles in both reproductive tract and nerve growth, we decided t...
